Lactobacillus acidophilus – one of the dominant vaginal microorganisms of a healthy woman, with antagonistic activity against pathogenic and conditionally pathogenic microorganisms.
Estriol – is a female sex hormone, short-acting estrogen. It has a protective trophic effect on the epithelium of the vagina and does not have a systemic effect on the body of a woman. The proliferating mature vaginal epithelium accumulates glycogen, necessary for the functioning of lactobacilli, which, in turn, produce lactic acid, supporting the acidic environment of the vagina (pH 3.8-4.5), which creates unfavorable conditions for the colonization and development of pathogenic microorganisms. In addition to lactic acid, acidophilic lactobacilli produce hydrogen peroxide and bacteriocins, which also inhibit the development of pathogenic and conditionally pathogenic microorganisms.
A change in the physiological flora of the vagina occurs due to various factors: local or systemic use of antibacterial drugs, including due to vaginal infections, systemic diseases, non-compliance with hygiene rules. In these cases, the number of lactobacilli can be significantly reduced. The intravaginal use of the drug Ginoflor E improves the condition of the vaginal epithelium and restores its normal flora. Lactose, which is contained in vaginal tablets as an excipient, can also be fermented by lactobacilli into lactic acid.
The vaginal epithelium becomes thinner and can be damaged even with hormonal disorders, and therefore Ginoflor E can be used in postmenopausal women to relieve its local symptoms (itching, burning, dry vagina, soreness during intercourse), helping to normalize the state of the vaginal and urethral epithelium. , reducing the incidence of dystrophic inflammatory conditions.
In vitro, it has been shown that acidophilic lactobacilli within a few hours lead to the necessary decrease in pH. Estriol, with a treatment duration of 6-12 days, provides a proliferative trophic effect on the vaginal epithelium.
Pharmacokinetics
When the drug is injected into the vagina, lyophilized bacteria and estriol are released.
Absorption of estriol from Gynoflor E has been studied in postmenopausal women. After repeated use of the drug, the concentration of estriol in plasma corresponds to the concentration of endogenous unbound estriol. After the 12th day of the use of vaginal tablets during treatment with Ginoflor E (1 time per day), the maximum concentration of unbound estriol in the blood plasma does not differ from the initial one, which indicates the absence of systemic absorption.
The use of the drug does not affect the plasma concentration of sex hormones present in the body of a woman – estrone and estradiol, since estriol is the final product of their metabolism.
Indications
Restoring normal vaginal microflora after topical and / or systemic use of antibiotics or antimicrobials (including vaginal infections: bacterial vaginosis, non-specific vulvovaginitis of urogenital infections and sexually transmitted diseases).
Post-menopausal estrogen-dependent atrophic vaginitis, including in combination with hormone replacement therapy (HRT).
Contraindications

Use during pregnancy and lactation
Use in the first trimester of pregnancy is undesirable. Perhaps the use of the drug Ginoflor E in the II-III trimesters of pregnancy and during breastfeeding. Undesirable effects of lactobacilli and estriol on pregnancy and the condition of the fetus / newborn were not found. However, long-term clinical studies on the effect of estriol on the fetus have not been conducted.
Dosage and administration
Vaginal tablets must be administered deep into the vagina in the evening before bedtime, preferably in the supine position, slightly bending the legs at the knees.
To restore normal vaginal microflora after topical and / or systemic use of antibiotics or other antimicrobials (including vaginal infections: bacterial vaginosis, non-specific vulvovaginitis, urogenital infections and sexually transmitted diseases): 1-2 vaginal tablets daily for for 6-12 days.
In the treatment of estrogen-dependent atrophic vaginitis in postmenopausal women: 1 vaginal tablet daily for 6-12 days, then the maintenance dose is 1 vaginal tablet 1-2 times a week.
Side effects
Possible sensation of burning (heat) in the vagina, rarely redness and itching of the vulva and vagina.
