Drug Interaction
The risk of developing hepatotoxic action of paracetamol is increased by the simultaneous administration of barbiturates, diphenin, carbamazepine, rifampicin, zidovudine and other inducers of microsomal liver enzymes. Enhances the effects of sedatives, ethanol, monoamine oxidase inhibitors. Antidepressants, phenothiazine derivatives, antiparkinsonian and antipsychotic medicines increase the risk of urinary retention, dry mouth, constipation. Glucocorticosteroids increase the risk of developing glaucoma. Paracetamol reduces the effectiveness of uricosuric drugs. Chlorophenamine together with furazolidone can lead to hypertensive crisis, agitation, hyperpyrexia. Tricyclic antidepressants increase the adrenomimetic effect of phenylephrine, and the simultaneous administration of halothane increases the risk of ventricular arrhythmia. Reduces the antihypertensive effect of guanethidine, which in turn enhances the -adrenostimulating activity of phenylephrine. With long-term regular use of paracetamol, the anticoagulation action of warfarin and other coumarin drugs is increased and the risk of bleeding is increased.
Overdose
Symptoms caused by paracetamol are manifested after ingestion of more than 10-15 g: pale skin, decreased appetite, nausea, vomiting, hepatonecrosis, increased activity of hepatic transaminases, increased time of prothrombin.
Treatment: gastric lavage in the first 6 h, administration of SH-group donors and precursors of glutathione-methionine synthesis 8-9 h after overdose and N-acetylcysteine after 12 h. In case of accidental overdose, see a doctor immediately any symptoms of overdose or not.
